食管鳞癌中 LSD1 与 Notch 靶基因启动子结合调控 Notch 和 PI3K/Akt/mTOR 通路

摘要:LSD1Lysine-specific demethylase 1),Notch 和 PI3K/Akt/mTOR 信号通路在多种癌症中处于异常活化的状态,包括食管鳞癌,但 LSD1 对 Notch 和 PI3K/Akt/mTOR 信号通路的调控关系目前还不清楚。本研究主要探讨食管鳞癌中 LSD1 对 Notch 和 PI3K/Akt/mTOR 信号通路的调控关系及机制。首先我们证实 LSD1Notch 和 PI3K/Akt/mTOR 信号通路相关蛋白在食管鳞癌细胞中存在表达。其次,我们发现用 TCP 或者 LSD1 shRNA 抑制食管鳞癌细胞中 LSD1 的表达后能够降低 Notch 和 PI3K/Akt/mTOR 信号通路相关蛋白的表达。最后,我们证实 TCP 处理食管鳞癌细胞后可抑制 LSD1 与 Notch3Hes1CR2 启动子区域的结合。
结论:食管鳞癌中 
LSD1 可能通过结合 Notch 通路相关基因的启动子区域来正向调控 Notch 和 PI3K/Akt/mTOR 信号通路。
