摘要:线粒体功能障碍是多种疾病,尤其是中枢神经系统疾病的关键病理机制之一。因此,能够有效恢复线粒体功能的治疗策略在中枢神经系统疾病中展现出巨大潜力。纳米技术已成为这一领域的重要平台;凭借其可修饰性、可控性和靶向能力,纳米系统能够实现在线粒体层面的精准干预。本文综述了线粒体功能障碍在中枢神经系统生理及病理过程中的核心作用,并系统总结了关键的线粒体靶向策略,包括小分子配体、线粒体靶向肽、线粒体膜衍生囊泡以及仿生膜涂层技术。同时,我们探讨了线粒体靶向纳米系统在主要中枢神经系统疾病中改善线粒体功能障碍的疗效,涵盖神经退行性疾病、脑肿瘤、缺血性卒中及创伤性脑损伤等代表性疾病。最后,本文指出了当前线粒体纳米医学在转化应用中面临的挑战及推动其未来发展的关键研究方向。总体而言,本综述系统梳理了线粒体纳米治疗领域的最新进展,强调了其在治疗中枢神经系统疾病中的应用潜力。
