血清miR-377-3p的异常表达及其在颈动脉狭窄中的诊断价值

摘要  

目的:明确颈动脉狭窄(CAS)患者血清中miR-377-3p的表达特征,并评估其临床应用诊断价值。  

患者与方法:采用qRT-PCR技术检测78例CAS患者与78例匹配健康对照者血清样本中miR-377-3p的表达水平。通过Pearson相关性分析指标间关联,利用ROC曲线评估miR-377-3p的诊断效能。同时对人主动脉平滑肌细胞(HASMCs)的细胞功能进行检测,并通过荧光素酶报告基因实验对生物信息学预测的靶向调控关系进行验证。

结果:诊断为CAS的患者血清miR-377-3p水平降低。基于ROC曲线的分析显示,miR-377-3p具有较高的诊断价值,曲线下面积(AUC)为0.9597。血清miR-377-3p与收缩压(SBP)、舒张压(DBP)、空腹血糖(FBG)和低密度脂蛋白胆固醇(LDL)呈负相关,而与高密度脂蛋白胆固醇(HDL)呈正相关。miR-377-3p可抑制HASMCs的增殖和迁移,但EDIL3能够抵消这种效应。

结论:这些结果表明,miR-377-3p在CAS管理中具有双重作用,既是潜在的生物标志物,也是可能的治疗靶点。miR-377-3p可通过靶向EDIL3抑制HASMCs的增殖和迁移。
