摘要 泛凋亡(PANoptosis)是一种近年来提出的新型程序性细胞死亡方式,其特征是由多蛋白复合物泛凋亡小体(PANoptosome)共同调控细胞焦亡、凋亡和坏死性凋亡三种经典死亡途径,且无法被其中任何一种途径单独解释。泛凋亡的启动依赖于泛凋亡小体的组装与激活,该过程已被证实与炎症性疾病、神经退行性疾病、癌症及肺部疾病等多种疾病的发生发展密切相关。然而,泛凋亡在药物性肝损伤(DILI)中的作用尚未得到全面的讨论。DILI是临床常见的药物不良反应,其发生与肝细胞死亡密切相关。因此,本文旨在综述泛凋亡的分子机制及其泛凋亡小体的组装过程,探讨细胞凋亡、坏死性凋亡和焦亡之间的相互作用,并深入分析泛凋亡在DILI中的潜在作用。深入探索天然产物通过抑制泛凋亡小体的组装,或靶向ZBP1AIM2NLRP3等关键节点以干预泛凋亡,有可能为治疗DILI提供巨大机遇。
