摘要:基因组学与分子生物学的最新进展加速了基于核酸的治疗药物的开发。为优化其临床疗效,研究者们探索了多种递送系统,包括脂质纳米粒、聚合物纳米材料、无机纳米颗粒、细胞外囊泡、病毒载体、蛋白质载体以及核酸偶联物,旨在提高核酸药物的稳定性、细胞摄取率和生物利用度。本文综述了核酸治疗药物的现状,重点阐述其分类、递送策略以及在代谢性和炎症性疾病中的应用。文章着重分析了这些递送系统的作用机制、靶向递送方法以及当前面临的挑战。文献检索自PubMed和Web of Science数据库,筛选标准包括与主题的相关性及发表时间(2020-2025年)。本文旨在为优化核酸药物及开发新型递送系统提供见解,为复杂疾病的治疗提供新的视角。
